A study on the Cloning of a new member of tumor necrosis factor receptor-associated protein (nTRAF)

Abstract
Several ring finger proteins that are associated with the cytoplasmic tails of members of tumor necrosis factor receptor(TNFR) superfamily have been identified. Because the TNFR-associated factors(TRAF) appear to be physically associated with the receptors, they are most likely to initiate receptor-mediated signaling.
To date, three of such proteins are reported, named TRAP-1, TRAF-2 and TRAF-3(or CD40bp, CRAF-1, LAP-1). This study identified the fourth member of TRAF family called nTRAF from Expression sequence tag. The nTRAF which is composed of 470 amino acid is 40-50% similar to the other TRAF amino acid sequences. nTRAF contains a N-terminal ring finger domain and seven repeats of zinc finger motif, and a C-terminal TRAF domain, but does not contain isoleucine zipper domain. nTRAF-expressing mRNA is expressed ubiquitously in various cell lines, especially in Burkitt¢¥s lymphoma Raji, and chronic myelogenous leukemia K-562.
