Effects of Raloxifene on Cognitive Function in Postmenopausal Women: Meta-Analysis of Randomized-Controlled Trials

Abstract
Background: Raloxifene has been widely used for the treatment of osteoporosis in postmenopausal women. However, its effect on cognitive function remains controversial. The aim of our study was to perform a meta-analysis to evaluate the effect of raloxifene on cognitive function in postmenopausal women.

Methods: We searched electronic databases including Pubmed, SCOPUS, Cochrane Library, and CINAHL from January 1990 to September 2016. A random-effects meta-analysis was performed with randomized-controlled trials to evaluate the effect of raloxifene on cognitive function in postmenopausal women. In addition, subgroup analyses were performed based on the dosage of raloxifene, study duration, quality of the studies, and the tool accessing cognitive function. The standardized mean differences of cognitive function tests were analyzed.

Results: Out of 359 searched studies, only 7 (4 studies with raloxifene 60 mg, 1 study with raloxifene 120 mg, 2 studies with both 60 mg and 120 mg) were included in the final analysis, all of which were randomized-controlled trials. The point estimate¡¾standard error was 0.084¡¾0.099 (P=0.394, I2=64.869%; 6 studies) for raloxifene 60 mg, and 0.004¡¾0.028 (P=0.883, I2¡´0.001%; 3 studies) for raloxifene 120 mg. Subgroup analysis by study duration or quality of the studies showed no significant differences. Raloxifene seemed to have a better effect on short-term memory (0.288¡¾0.261, P=0.269) than on long-term memory (0.150¡¾0.115, P=0.189), but there was no statistical significance.

Conclusion: This meta-analysis found that raloxifene may have little or no effect on cognitive function in postmenopausal women. Further studies are necessary to verify the effect of raloxifene.
