Effects of Acupuncture and Radix Astragali Aqua-acupuncture at Synsu(BL23) on Transcriptional Expression of Mouse Cytokine IL-4

Acupuncture and Radix Astragali aqua-acupuncture stimuli have many been used to cure human diseases. However, it still remains to be unknown on its action mechanism, physiolosical and biochemical aspects. Thus, many attempts were made to show the scientific background covering the above mentioned mechanisms. Most recent studies show that these tests improve blood circulatory system and increase leucocyte counts. In this study, we have applied the acupuncture stimuli to mouse Synsu(BL23), which is a stimulative point of oriental medicine, to see if cytokine such as IL-4 can be detected. Mice were treated with lipopolysaccharide(LPS) for inflammation induction, and then reverse transcriptase-polymerase chain reaction (RT-PCR) using each primer set was performed to trace the amounts of mRNA. The results are summarized as follow ; 1.IL-4 was expressed in LPS-nontreated mouse at 30min after acupuncture-stimuli, however, hardly expressed in after 1, 2hrs. 2.IL-4 was continuously expressed in LPS-treated mouse at 30min to 3hrs after acupuncture-stimuli, especially highly expressed at 30 to 60min. And IL-4 was a little decreased after 3hrs but was continuosly expressed. 3.IL-4 was highly expressed in LPS-nontreated mouse at 15min after Radix Astragali aqua-acupuncture stimuli, and expression of IL-4 gradually decreased at 30min to 3hrs. 4.IL-4 was gradually expressed in LPS-treated mouse at 30min to 3hrs after Radix Astragali aqua-acupuncture stimuli, especially highly expressed after 1hr.
Therefore, IL-4 in LPS-treated mouse was expressed more effectively than that of LPS-nontreated mouse after acupuncture and Radix Astragali aqua-acupuncture stimuli. And in LPS-treated mouse IL-4 after Radix Astragali aqua-acupuncture stimuli was expressed more effectively than after acupuncture stimuli. On the other hand, now, we are in progress to elucidate the immunological action mechanism of acupuncture and Radix Astragali aqua-acupuncture stimuli. And cytokine IL-4 can be used not only basis of the effect of acupuncture and Radix Astragali aqua-acupuncture but a diagnostic guide through the immunological actions of those.
