In vitro chromosome aberration test of CA-YAKCHIM using mammalian cultured cell

Abstract
Objectives : The purpose of this study was to investigate the chromosomal aberrations induced by CA-YAKCHIM in Chinese Hamster Lung cells.

Methods : In order to determine the high dose levels of the main study, the growth inhibition study was performed first, The high dose was selected at 5,000 ¥ìg/mL and it was sequentially diluted to produce lower dose levels of 2,500, 1,000, 500, 250, 100, 50, 10, and 5 ¥ìg/mL.

Results : As a result, cytotoxicity and deposition by the CA-YAKCHIM were not evident at any dose level in the short time treatments without and with metabolic activation and the continuous treatment without metabolic activation. Therefore, the dose levels of the main study were selected as 1,250, 2500, and 5,000 ¥ìg/mL. In the main study, the frequency of cells with structural chromosome aberrations was less than 5% in the short time treatments with and without metabolic activation and the continuous treatment without metabolic activation. However, frequency of cells with numerical chromosome aberrations was more than 5% in the continuous treatment with and without metabolic activation.
Conclusions : In this study, CA-YAKCHIM did not exhibit structural chromosome aberrations. However CA-YAKCHIM exhibited numerical chromosome aberrations. Thus, for the more adequate evaluation of the genotoxic potential of CA-YAKCHIM, other in vivo genotoxicity test is needed.
