Mapping Schizophrenia Genes by Linkage Analysis

Data from genetic epidemiological studies have indicated that genetic factors play a major role in determining susceptibility to schizophrenia. Linkage analysis in multiplex families with schizophrenia is a crucial step in the investigation of the susceptibility genes of the disease. Even though genome-wide linkage scans have produced conflicting results, modest evidence of linkage with schizophrenia has been reported for several chromosomal loci (1q32-42, 6p24-22, 8p21-12, 13q32-34, 22q11-22, etc). In addition, some promising positional candidate genes have been identified (DISC1, DTNBP1, NRG1, DAO(A)(G72/G30), COMT, etc). This review summarizes study designs and methods, and recent results of genome-wide linkage studies of schizophrenia. We discussed on main hurdles and points at issues in this research field and future perspective of gene mapping studies of schizophrenia. Further efforts are needed to increase statistical power to identify causative alleles each exerting a small to moderate effect on the overall disease risk. Various approaches to validate clinical and biological phenotypes for schizophrenia are also warranted.
