Effect of PGF3¥á on the Survival and Production of Nitric Oxide in Cultured Trabecular Meshwork Cells

Abstract
Purpose: To investigate whether prostaglandin F2¥á(PGF2¥á) affect on the survival and production of nitric oxide
(NO) in cultured human trabecular meshwork (TM) cells.

Methods: Primarily cultured human TM cells were exposed to 0, 100, 200, 500 nm PGF2¥á(PhXA85, latanoprost free acid) using serum-deprived media for 3 days to evaluate the effect on the cellular survival and nitric oxide production assessed by MTT and Griess assay, respectively. Also evaluated was in hypoxic conditions induced by sodium cyanide.

Result: PGF2¥ásignificantly decreased cellular survival of cultured TM cells in a dose-dependent manner accompanied with decreased NO production. PGF2¥áwas not cytoprotective in hypoxic conditions.

Conclusion: The current results suggest that PGF2¥ádecreased cellular survival and production of NO. This PGF2¥á-induced decreased NO production in TM cells may be involved in the down-regulation of trabecular outflow.
