Effect of Brimonidine on the Survival and Production of Nitric Oxide in Cultured Trabecular Meshwork Cells

Abstract
Purpose: To investigate whether brimonidine affect on the survival and production of nitric oxide (NO) in cultured
human trabecular meshwork (TM) cells.

Material and Methods: Primarily cultured human TM cells were exposed to 0, 10, 50, 100 ¥ìM brimonidine using 1% serum-containing media for 1 day and 3 days. The cellular survival and nitrite production were assessed with MTT and Griess assay, respectively.

Results: Cellular survival of cultured TM cells was significantly decreased after exposure for 1 day and 3 days to 100¥ìM brimonidine. Production of NO was decreased after exposure for 1 day and 3 days to 50 ¥ìM and 100 ¥ìM brimonidine, respectively. Production of NO was decreased after exposure for 3 days to 100 ¥ìM brimonidine.

Conclusion: The current results suggest that brimonidine decreased cellular survival and production of NO. Thus, brimonidine could decrease aqueous outflow through trabecular meshwork.
