Morphological Changes of DBA- and VIP-Positive Taste Cells in Rat Circumvallate Taste Buds after Glossopharyngeal Axotomy

Abstract
Purpose: The present study was carried out to investigate the structure and the reactivities of dolichos biflorus agglutinin (DBA) and vasoactive intestinal polypeptide (VIP) of degenerating taste buds in the circumvallate papilla.

Materials and Methods: The bilateral glossopharyngeal nerves of the male Sprague-Dawley rats (250~300 g) were cut and these animals were sacrificed with time after at 1, 3, 5, 7, 10, 15 and 30 days. The circumvallate papilla of the
tongue was dissected and then the tissues were fixed by using of 4% paraformaldehyde solution. Tissue samples cut
into 10 ¥ìm thickness on the cryostat and were immunohistochemically stained for the DBA and VIP.

Results: The number of the taste buds in the circumvallate papilla was not changed in the control group to 30 days group, however, the taste buds and taste cells were atrophied from 3 days group and the vacuoles were appeared in
the nuclei of the taste cells from 7 days group. In the control group, the number of DBA- or VIP-positive taste cells were observed about 10 or 3~4 in the taste buds, respectively. From double labelling method, DBA- or VIP-positive taste cells were different. The reactivities of DBA-positive taste cells were similar from the control group to 5 days group, however, the reactivities were decreased from 7 days group and kept to 30 days group. The reactivities of VIPpositive taste cells were similar from the control group to 7 days group, however, the reactivities were disappeared from 10 days group.

Conclusion: These results suggest that DBA- or VIP-positive taste cells are different taste cells and the degeneration time of these cells are different.
