An Immunocytochemical Study on the Substance P-ergic Neurons in the Olfactory Bulb of the Cat


Abstract
Substance P is an undecaapeptide, chemically characterized by Chang and Leeman(1970), which is thought to act as a neurotransmitter or neuromodulator in the mammalian central nervous system. It has been shown by immunocytochemical techniques
using
substance P antiserum that substance P-ergic neurons are widely distributed in various regions of the brain including the olfactory bulb. Previous immunocytochemical studies have provided the evidence for the presence and identification of
substance
P-ergic neurons in the olfactory bulb of rodents light microscopically(Baker, 1986 ; Matsutani et aL, 1089). However, little ultrastructural studies have been done to clarify the synaptic organization of substance P-ergic neurons in the olfactory
bulb.
This study was conducted to identify the substance P-ergic neurons and to define their synaptic cirsuitry in the olfactory bulb of the cat by pre-embedding immunocytochemical method using substance Pantiserum light and electron microscopically.
@ES The results were as follows :
@EN 1. Substance P-ergic neurons of the cat olfactory bulb were some granule cells located in the granule cell layer and a few external tufted cells located in the periglomerular region.
2. The processes of substance P-ergic granule cells made synapses with soma and the proceses of non-substance P immunoreactive granule cells and dendrites of mitral cells.
3. Dendrites of substance P-ergic tufted cells had input synapses from non-substance P immunoreactive axon terminals of the olfactory nerves within the giomeruli.
The results demonstrate that substance P-ergic granule cells make synapses with non-substance P immunoreactive granule and mitral cells, and substance P-ergic tufted cells, with non-substance Pimmunoreactive olfactory axon terminals. and they
might
play
an important role in the processing of olfactory informations in the olfactory bulb.
