Comparison of the Effects Between Bevacizumab and Mitomycin C on the Survival of Fibroblasts

Abstract

|
|
Purpose: To compare the antiproliferative effects between bevacizumab, a monoclonal antibody against vascular endothelial growth factor (VEGF), and mitomycin C in human Tenon¡¯s capsule fibroblasts.
Methods: Primarily cultured human Tenon¡¯s capsule fibroblasts were exposed to 0, 25, 50, 100, and 200 ¥ìg/ml bevacizumab and mitomycin C, and incubated for 5 days. Cellular survival and production of nitric oxide were assessed by MTT assay and Griess assay, respectively.
Results: Bevacizumab showed antiproliferative effects only at high concentrations (200 ¥ìg/ml) and revealed much less effect on the cellular survival compared to mitomycin C. In addition, bevacizumab did not affect the production of nitric oxide.
Conclusions: The antiproliferative effect of bevacizumab is much lower than mitomycin C in human Tenon¡¯s capsule fibroblasts.
