Abstract
This experiment was observed the effect of organophosphorus pesticides on the secretions and productions of the mucosubstances in the duodenal glands and goblet cells of the duodenal mucosae by histochemical methods.
Healthy adult rats weighing about 250-300g each were divided intotwo groups-normal and experimental. Organophosphorus pesticides used in this study were DDVP (35mg/kg), malat¡þhion (2.5g/kg), dipterex (200mg/kg) and EPN (25mg/kg),¡Æ and administrated to the experi¡þmental groups by orally. They were sacrificed on the 3, 6, 12, 24 and 48 hours after the administration. The mucosu bstances were stained with alcian blue pH 2.5, periodic acid Schiff (PAS), alcian blue pH 2.5-PAS, alcian blue pH 1.0-PAS and aldehe chsin pHI.7¡þalcian blue pH 2.5 method.
The obtained results were as follows
1. In the normal group, the duodenal glands contained large amount of neutral mucin. But duodenal goblet cells contained abundant acid and neutral mucins, and the former was more than the latter.
There were two types of goblet cells; ones sulfated and neutral mucins and others nonsulfated and neutral ones. The latters were much more than the formers.
2. The amount of the neutral mucins in the duodenal glands were a little different among rats administered by the Kinds of pesticides, but they were remarkably decreased from 3 hours to 12 hours after the administration.
The diminution of neutral mucins were more remarkable in dipterex and EPN administra¡þtion groups than in DDVP and malathion groups. The diminished neutral mucin in experimental
groups started to recover from 24 hours after the administration of pesticides.
3. The amounts of acid and neutral mucins in goblet cells were remarkably diminished in 3 and 6 hours after the administration, and the diminution of acid mucins was more remarkable than that of neutral ones. But they were recovered from 12 or 24 hours after the administration.
The mucins were more decreased in rats given the DDVP, dipterex and EPN than malat¡þhion, and the mucins of goblet cells in upper villi and lower crypts were more decreased than lower villi and upper crypts.
The number of goblet cells which secrete sulfated mucins was decreased from 3 to 12 hours after the administration, and then they have a tendency to increase from 24 hours. Those which secrete nonsulfated mucins were increased from 6 to 12 hours after the administration, but they tend to decrease from 24 hours after the administration.
On the basis of the above results, it is suggested that the mucosubstances were decreased from early period after organophosphorus pesticides administration. Furthermore, the pro¡þduction of acid mucins, especially the metabolism of sulfated mucin, seems to be inhibited more than neutral ones.
