ABSTRACT- An HPLC procedure with UV detection has been developed for the quantitation of biphenyl dimethyl dicarboxylate(DDB) in human plasma. The drug and the internal standard(diazepam) were extracted from plasma with ether and quantitated using a reverse-phase C_18 column. The chromatograms were completely free from interfering peaks, and the retention times of DDB and the internal standard were 6.6 min and 8.9 min, respectively. Calibration plots were linear over the concentration range of 10 - 200 ng/ml of DDB with the correlation coefficients all higher than 0.998. The mean interday precision and accuracy over three days were 5.71% and 4.28%, respectively. The mean recoveries over the entire calibration range were 96.9% for the drug and 94.2% for the internal standard. The limit of detection for DDB was 0.5ng/ml. The method was simple, reliable and accurate for the quantitation of DDB in human plasma.
