Morphological Study of Nerve Fibers in the Stored Rabbit Cornea

Abstract
The corneas were excised with the 1-2mm scleral rim from adult rabbits under sterile conditions immediatedly after they were sacrificed. The corneas were preserved for the pre-determined period under various conditions described below and the morphological study of degenerative changes of the corneal nerve fibers were performed by modified Romanes¢¥ silver stain mithod after fixation in 10% neutral formalin solution and 10-15§Á paraffin serial section.
The results were as follows:
1. The degenerative changes of the verve jibers, more or less, could be soon observed on the first day in all corneas under all methods of preservation.
2. Optimum preservaton was attained when they were stored in liquid paraffin at 4¡É and in A.G. solution (mixture of 50% alcohol and 100% glycerin in equal amounts) tat room temperature respectively. In the specimens thus treated, the gradual degeneration and disappearance of nerve jibers could be continuously seen during the 40 week observationperiod)in liquid paraffin)and the 46 week period (in A.G. solution), the degenerated fine nerve fibers still remaining.
3. Most rapid and remarkable morphological changes and disapperance of nerve fibers were recognized in the specimens preserved in 20% and 40% glycerin at room temperature, and at the end of the 26 week period, the nerve fibers disappeared almost completely.
4. The changes of nerve fibers were almost similar both in the group preserved in normal saline solution and the one in 15% glycerin at 4¡É, in both of which the degenerated nerve fibers could be observed still remaining at the end of the 34 week period (in normal saline solution) and at the end of the 40 week period (in the 15% glycerin), though the degenerative change was more typical and remarkable in the normal saline solution series, where the globular nodules formation in the nerve fibers was distinct.
