Blood Pressure Responses to Acetylcholine and Dimethylphenylpiperazinium (DMPP) in the Rabbit

Abstract
Being different from actions in the cat and dog, acetylcholine and dimethylph-enylpiperazinium(DMPP) hardly produced pressor effect in the atropinized whole rabbit. Treatment of the rabbit with either bretylium(10mg/kg), guanethidine(10mg/kg)or reserpine (2mg/kg), which markedly potentiated pressor effect of epinephrine, could make acetycholine and DMPP elicit pressor effect. Cocaine, which potentiated the pressor effect of catecholamines in the cat and dog, did not affect the effects of catecholamines, acetylcholine and DMPP.
The pressor effect of acetycholine and DMPP in the rabbit treated with the above-mentioned agents was abolished or very much reduced by bilateral adrenalectomy, phenoxybenzamine, hezamethonium or P-286. There results indicated that the pressor action of acetylcholine and DMPP in the rabbit was due to catecholamines released mainly from adrenal medulla. Catecholamine contents of rabbit adrenal glands were not affected by treatment of guanethidine (10mg/kg, 2 hours later)but reduced to about half of tti by reserpine (2mg/kg,2.5 hours later).
In the atrpinized spinal rabbit DMPP and large doses of acetylcholine could produce pressor effect, which was abolished or markedly reduced by phenoxybenzamine, hexamethonium or P-286.
The possible mechanisms which distrurb inducement of the nocoinic effect of acetylcholine and DMPP in the rabbit blood pressure were discussed.
