Stop-flow Analysis of the Diuretic Action of Ethacrynic acid in the Dog

Abstract
The diuretic action of a new potent diuretic, Ethacrynic acid[2,3-dichloro-4-(2-methylenebutyryl)-phenoxy)-acetic acid, was analysed , utilizing the stop-flow technique in the dog.
Ethacrynic acid depressed the reabsorption of sodium ion in the proximal and distal tubules, as well as in the ascending limb of the Henle¢¥s loop, resulting in marked natriuresis and diuresis, and in the decrease of the concentrating ability of the kidney.
It enhances the potassium excretion through the kidney, has no influence upon the active transport of PAH, and inhibits the acid secretion in the distalportion of the tubules.
The drug can unfold its full effect even under the maximum action of a mercurial diuretic, Mersalyl, thus suggesting a different mechanism from that of organomercurials.
