The Inhibition of Erythrocyte NAD Nucleosidase by Porphyrin Derivatives

Abstract
Hematin, protoporphyrin and bilirubin were found to be potent inhibitors of NAD nueleosidase in rabbit mature erythrocytes membrane. The concentration which caused 50% inhibition was 0.3mM for hematin and 0.6 mM for protoporphyrin. Rabbit spleen and brain NADases were also inhibited by hematin but to a lesser degree.
The inhibition by hematin was found to be of an noncompetitive type and was prevented by the Presence of human globin, bovine serum albumin.and-human serum proteins. However, when bovine serum albumin was added after the addition of hematin to the NADase solution, the inhibition by hematin was not abolished.
Zinc ion which repartedly binds to the histidyl residues in human serum albumin could not give any influence upon the activity of the erythrocyte NADase or on the inhibition by hematin.
From these result it was suggested that the inhibition of NADase by bematin might be might be caused by the binding of hematin to, NADase through its porphyrin and that hematin NADase, complex is essentially undissociable.
