Effect of Testosterone and Estrogen on the Levator Ani muscle of the Rat

Abstract
This experiment was designed for the evaluation of the effects of testosterone and estrogen on the skeletal muscle.
Levator an imuscle sensitive to testosterone were evaluated by muscle mass index (mg muscle/g body weight; MMI) and muscle fiber size following the treatment of testosterone or estrogen respectively to the normal and castrated rats.
Histological and enzyme histochemical changes were also evaluated by H-E,
trichrome, PAS, succinate dehydrogenase (SDH), adenylate deaminase (ADA), alkaline phosphatase, acid phosphatase and myosin-ATPase stains.
The results obtained were as follows.
1. Hypertrophy of Levator ani muscle was prominent in both normal and castrated rat groups, following the testosterone treatment.
2. In Levator ani muscie, the number of type IIA fibers and the enzyme activities of SDH, ADA and myosin-ATPase were far less and week compared with those of Tibialis anterior muscle.
3. The number of type IIA fibers and enzyme activities of SDH, ADA and myosin-ATPase were markedly reduced in Levator ani muscle, compared to those of Tibialis anterior muscle.
4. Activities of SDH and myosin-ATPase seemed to be increased in normal rats, following the testosterone treatment, but did not increase in castrated rats. SDH and myosin-ATPase activities were increased in the atrophied muscle fibers following the estrogen treatment.
5. Hypertrophy or atrophy of muscle fibers in Levator ani was evenly shown in all muscle fiber types, not limited to specific muscle fiber ones following the testosterone or estrogen treatment, respectively.
