Electron Microscopy on Glucose-6-phosphatase Activity in Hepatic Cells of Methotrexate Treated Rat



Abstract
Toxic effect of methotrexate on glucose-6-phosphatase activity in rat hepatocytes was studied by electron microscopy and electron microscope cytochemistry. Rats received methotrexate every other day for 13 times intraperitoneally and were sacrificed 24 hours after last injection of the drug. Liver tissues were prepared for electron microscope cytochemistry of glucose-6-phosphatase and were observed with electron microscope. Results obtained were summarized as follows.
Hepatocytes from the rats treated with methotrexate revealed marked proliferation of smooth endoplasmic reticulum, a depletion of glycogen particles and a increase of fat droplets.
In electron microscope cytochemistry, glucose -6-phosphatase activity in normal rat hepatocytes was localized to the rough and smooth endoplasmic reticulum and the-nuclear envelope. These localization of the enzyme was not altered in the hepatocytes of the experimental animals. However, enzyme activity was noted to be, generally, diminished.
These results suggest that methotrexate administration give rise to an enhanced synthesis of smooth endoplasmic reticulum, but inhibit or does not appreciably affect synthesis of glucose-6-phosphatase.
