The Generation of Superoxide Radical During the Autoxidation of Ascorbate

Abstract
The generation of superoxide radical during the autoxidation of ascorbate has been suggested in some reports, but none of them has demonstrated its presence unequivocally. In the present study, attempts were made to demonstrate the generation of superoxide in the autoxidation of ascorbate by examining the effects of ferrous ion, superoxide dismutase, catalase and peroxides on the rates of ascorbate oxidation and co-reduction of nitroblue tetrazolium (NBT) .
Ferrous ion enhanced the rate of ascorbate oxidation but inhibited the NBT reduction during the autoxidation of ascorbate. Both the rate of ascorbate oxidation and co-reduction of NBT were inhibited by superoxide dismutase, catalase and peroxides.
The hydroxyl radical scavengers such as ethanol and mannitol partly inhibited the co-reduction of NBT.
Based on these results, the generation of superoxide radical and hydrogen peroxide during the autoxidation of ascorbate is discussed, and a reaction mechanism is proposed.
