The Effect of Nitrogen Mustard on Hair Cells of the Organ of Corti

Abstract
To improve the treatment of malignant tumor, many chemotherapeutic agents have been invented and widely used. Nitrogen mustard, one of the alkylating agents is the chemical agent used in the treatment of lymphocytic leukemia and Hodgkin¢¥s disease, and is known to have ototoxic effect. However the exact site and mechanism of the toxic effect on the hair cells of the organ of Corti are uncertain. Cytological observation was carried out on the hair cells after administration of cyclophosphamide. (a kind of nitrogen mustard. )
Healthy guinea pigs were treated with cyclophosphamide daily in a dose of 5mg/kg B. W. for one week orally and were sacrificed, and fixed with 1 % osmic acid. Morphological findings of the hair cells were observed under the surface preparation methods of cochlea.
The results were as follows;
1. Cyclophosphamide produces most severe and significant changes in the basal turn, less severe in the 2nd turn and mild in the apical turn of the cochlea.
2. Cellular disarrangement is most severe in the whole of the basal turn and the lower part of the second turn of the cochlea.
3. The nuclei of the outer hair cell markedly changed than the inner hair cells.
