Studies on the Impairment of Liver Function and the Changes of Coagulation Factors due to Brontobenzene


Abstract
It is a well known fact that the liver is the most important organ which is related to the production of blood coagulation factors. Thus, there are many reports on the effects of various hepatotoxic substances on the blood coagulation, nevertheless, little has been known on the effect of bromobenzene without its toxic action to the hepatic cells.
Therefore, the author investigated the deficiencies of blood coagulation factors, hepatic functional disturbances and histological changes in liver of rabbits intoxicated with bromobenzen experimentally.
Albino rabbits weighing approximately 2 kg. were used as experimental animals. Four ml. of 20% bromobenzene in corn oil were injected intramuscularly one or two times.
The results were summarized as follows:
1) In the group of rabbits received injection of bromobenzene one time, the liver tissues showed the cloudy swelling of parenchymal cells and centrolobular fatty changes. There were no significant changes in liver function tests except for remarkably increased serum cholesterol level and transaminase activities. Blood coagulation time was moderately prolonged and the thromboplastin generating activity in serum was depressed but the depression of stable and labile factors and prothrombin were only slight.
2) In the group of rabbits received bromobenzene two times, the morphological changes of the liver were more advanced and centrolobular necrotic areas occurred. Liver function test also showed an increase in the amount of serum cholesterol and the activity of serum transaminase, however, the other tests remained unchanged. The prolongation of blood coagulation time was more remarkable, and the activities of labile factor and thromboplastin generation in serum were depressed and prothrombin was only slightly prolonged.
