Cytological studies of spermatogenesis in Bombina orientalis

Abstract
In the Bombina orientalis, the testis were observed by means of light and electron microscopes.
1. The results by light microscope were as follows:
a. The testis of Bombina orientalis consisted of many seminiferous tubules which were surrounded by interstitial tissues.
b. The seminiferous tubules of Bombina orientalis consisted of 5 kinds of cells arranged from the basal layer to lumen; the spermatogonia at the basal layer, the primary spermatocyst, the secondary spermatocyst and the spermatid cyst which were round or oval shapes, and the sperms which filled the empty spaces of the lumen or the space between the cysts.
c. Especially, the sertoli cells were found in only one side or either side of spermatogonia.
2. The results by electron microscope were as follows:
a. The primordial germ cells contained a large and highly lobate nucleus and one or two conspicuous nucleolus were observed.
b. The nucleus of the spermatogonia had one nucleolus and the shape of oval. A kind of remarkably granular material was observed in the cytoplasm.
c. Synaptonemal complex was found in the nucleus of the primary spermatocyte.
d. The secondary spermatocyte was much smaller than the primary spermatocyte, and the cytoplasm of the secondary spermatocyte became an oval shape with one side narrower than the other. Also, the intercellular bridge which connected individual cells was observed.
e. The nucleus of the spermatid was filled with irregularly condensed chromatin granules, and the acrosomal vacuole appeared in the cytoplasm.
f. The sperms which were divided into three parts were seen in the lumen; the head with a nucleus, the mid-piece in which the mitochondria were abundant and the tail consisting of undulating membrane attached around a flagellum.
