Studies on the therapeutic effects of Distocide^(®) in clonorchiasis

Abstract
|
|
|
In the clinical therapeutic trials, praziquantel, a product developed in joint research by E. Merck and Bayer AG proved to be highly effective against Clonorchis sinensis infection(Rim and Yoo, 197917)) .
Recently, the synthesis of praziquantel was succeeded by innovated technique in Korea, and it is already, in production. This. study was aimed to test the curability and to assess the side effects of Distocide® in human clonorchiasis.
A total of 85 patients with proven Clonorchis sinensis infection were treated with Distocide® at 3 dose levels. Twenty nine patients received a single dose of 40 mg/kg bwt(=body . weight), 28 patients received 2X30 mg/kg bwt one day and 28 patients were treated with 3 x 25 mg/kg bwt on a single day. Follow-up examinations were carried out at 3 weeks after treatment.
The results obtained in this study are as follows:
1. At a single dose of 40 mg/kg bwt, only 12 patients(41. 4 %) were parasitologicallycured and ERR (egg reduction rate) was 85.9 per cent.
2. Fifteen patients (53.6 %) out of 28 were cured with 2 x 30 mg/kg bwt and ERR was 96.8 per cent.
3. At a 3 x 25 mg/kg bwt, a recommend dose for clonorchiasis, 19 patients (67. 9 %) out of 28 were cured and ERR was 99.3 per cent.
4. Extended hematological and biochemical tests revealed no abnormal changes which could be induced by the compound after therapy.
5. Unwanted side effects such as headache and/or dizziness were mild and transient. Facial edema and urticaria appeared in 2 cases, but it was not the typical adverse effects of a domestic product.
6. The results suggest that praziquantel manufactured in Korea (Distocide?) is effective in the individual as well as mass treatment of human clonorchiasis.
