The Prophylactic Effect of Ketotifen in Bronchial Asthma


Abstract
Ketotifenn is a benzocycloheptathiophene derivative which has both anti-histaminic and antianaphylactic properties. Previously controlled clinical studies have established that this agent has prophylactic effect in bronchial asthma of children.
The purpose of this study is to evaluate the prophylactic effect in bronchial asthma of children. A study was carried out in 20 atopic asthmatic children who were 2 years to 15 years of age. Ketotifen was given orally at a dosage of 0.06 mg/kg/d twice daily for 12 weeks. The prophylactic. effect of this agent was assesed by asthma diary, opinion of parents and regular physical examination by physician.
The results were as follows:
1. Asthma attack scores, symptom scores, daily activity scores and sleep disturbance scores were decreased gradually during the course of treatment and markedly decreased after about 10th week of treatment. These findings were statistically significant (p < 0.05).
2. Concomitant medication was gradually decreased during the course of treatment and markedly decreased after about 10th week of treatment. This finding was statistically significant (p < 0.05).
3. Two cases complained mild drowsiness and 1 case complained mild headache but interruption of treatment was not needed. These side effects were subsided spontaneously within 1 month.
4. Asthma symptoms were improved in 14 cases (70%) of all patients at the end of study. The effects were rated as excellent in 3 cases, good in 7 cases, fair in 4 cases and no effect in 6 cases.
In conclusion, ketotifen appears to be effective and safe for the prophylaxis of bronchial asthma in children.
