Antioxidant and Anti-inflammatory Effects of Machilus thunbergii Leaves and Branch Extracts with Different Extraction Methods

Abstract
An ethnic plant of Machilus thunbergii (MT), belonging to the family Lauracecae, grows in warm areas of South Korea and Japan, China, and Philippines. In this study, we investigated the possible utilization of MT extracts from leaves and branch with different extraction methods for the investigation of antioxidant and anti-inflammatory activities. The leaves and branch of MT were treated with hot water, ultra-sonic cold water, 70% ethanol, and 100% ethanol to prepare the extracts. The extracts were analyzed for total phenolic and flavonoid contents as well as antioxidant properties such as DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging activity and 2,2-azino-bis (3-ethylbenzothiazoline- 6-sulfonic acid)diammonium salt (ABTS) radical scavenging activity. The highest total phenolic and flavonoid contents were found in the 100% ethanol fraction in branch and 70% ethanol fraction in leaves (311.38 ¡¾ 7.94 mg gallic acid equivalent (GAE)/g extract and 20.02 ¡¾ 2.45 mg Quercetin equivalent (QE)/g extract respectively). The 70% ethanol fraction and 100% ethanol fraction showed the strongest antioxidant activities in DPPH and ABTS assays. These results suggest that MT extract provides promising antioxidant and anti-inflammatory effects, and can be used as natural cosmetic substances in cosmetic industries in near future.
