Effect of Membrane Lipid Peroxidation on Liver Mitochondrial Functions


Abstract
The effect of the membrane lipid peroxidation levels on rat liver mitochondrial functions was investigated by in vitro test.
In in vitro test, the exposure time of liver homogenates to oxygen tensions was 0, 3, 6, 9, 12 hours and the lipid peroxidation levels were evaluated by the measurements fluorescent intensity and malondialdehyde(MDA). The fluorescent intensity of homogenates and mitochondrial suspension were elevated, and the concentration of MDA was not affected as time progressed. Therefore, fluorescent intensity for lipid peroxidation values were used to analize the correlation between lipid peroxidation levels and mitochondrial functions.
When the exposure time to oxygen was gradually increased in mitochondrial suspensions, fluorescent intensity was elevated, total mitochondrial activity and ATPase activity were decreased. The correlation between fluorescent intensity and total mitochondrial activity or ATPase activity were analyzed and the inverse correlation was -0.92, -0.64, respectively.
