Application of Multiplex PCR Assay for Detection of Methicillin-Resistant Staphylococcous aureus Isolated from Bovine Mastitis Milk






Abstract
37 of Staphylococcus aureus (S. aureus) and 33 of coagulase negative staphylococci (CNS) including 17 of methicillin resistant S. aureus (MRSA) and 15 of methicillin resistant coagulase negative staphylococci (MRCNS) strains isolated from bovine mastitis milk evaluated for detection of mec A gene by multiplex polymerase chain reaction (PCR) and broth dilution minimal inhibitory concentration (MIC). Multiplex PCR was carried out to detect the coa gene (pathogenic of Staph$ococcus aureus) and mec A gene (characteristically encoding for methicillin resistance in staphylococci). The mec A gene was found with 23 among 32 resistant strains, and m c A gene expression rate was higher in high MIC (¡Ã32§¶/ml) strains. All of the S. aureus isolates harbored the genes encoding staphylococcal coagulase. The total assay time was only about 1.5 hours when using multiplex PCR method applied in this study. Therefore, this method could be applied as a rapid technology for identification of MRSA among staphylococci.
