Inhibitory Effect of Reducing Agents and Amine Compounds on Glycation of Lysozyme

Abstract
The glycation of many proteins has been implicated as a major mechanism in the development of chronic diabetic complications as well as certain physical changes of aging. Reducing agents and amine compounds have been known as inhibitor in the glycation reaction. Simultaneous incubation with reducing agents and amine compounds inhibited the glycation of lysozyme with glycation initiators including fructose and ascorbic acid, glyoxal. These inhibitors prevented both the polymerization of lysozyme and the modification of amino acid residues of lysozyme. In the pre-incubation of reducing agents and amine compounds without lysozyme, the reaction with ascorbic acid was prevented but other reactions were not. The glycation initiator used in this work, therefore, may react with lysozyme in different manners. The identification of reducing agents and amine compounds as inhibitors of glycation of lysozyme suggests a potential inhibitory effect against the glycation in vivo.
