Identification of Glyoxal as the Reactive Intermediates during Autoxidative Glycosylation of Glucose and Fructose

Abstract
Glycation and autoxidation reactions contribute to protein modification in aging and diabetes. Formation of dicarbonyl compounds is the hypothetical first step in the glycation and subsequent browning of proteins. In order to identify the dicarbonyl compounds formed during autoxidative glycosylation of glucose and fructose under physiological conditions, glucose and fructose were incubated in 0.1 M phosphate buffer(pH 7.3) at 37¡É with lysine and/or transition metal chelators reagent. Dicarbonyl compounds were analyzed by spectrophotometry and HPLC after reaction with Girard-T reagent. The amount of dicarbonyl compounds increased with reaction time. HPLC analysis of adducts formed with Girard-T reagent indicated that glyoxal were the major ¥á-dicarbonyl compound formed during glycation reaction in vitro.
