This study was designed to investigate the feeding effect of docosahexaenoic acid (DHA) on lipid metabolism in brain of the learning and memory impairment animal model SAMP8. SAMP8 were fed basic diets(control group) with 15% of lard, and experimental diets(DHA group) with 5.0% of lard and 10% of docosahexaenoic acid (DHA)-rich fish oil (31.95% of DHA) for 8 months. Body weights gain in DHA group were consistently lower than those in control group, but there is no significant difference between these two groups. Grading scores of senescence and lipofuscin levels in DHA group were remarkably inhibited about 40% compared with control group. Total cholesterol of brain mitochondria and microsomes in DHA group were consistently 14% and 7% lower than those in control group. Docosahexaenoic acid (DHA) levels in DHA group were significantly increased about 14% compared with control group. These results suggest that grading score of senescence, total cholesterol and lipofuscin levels in brain of SAMP8 may be effectively inhibited by long-term administration of DHA-rich fish oil.
