Abstract
This study was designed to investigate the effects of docosahexaenoic acid (DHA)-rich fish oil on oxygen free radicals and their scavenger enzymes of learning and memory impairments SAMP8. SAMP8 were fed basic diets(control group) with 15% of lard, and experimental diets(DHA group) with 5.0% of lard and 10% of docosahexaenoic acid(DHA)-rich fish oil (31.95% of DHA) for 8 months. The passive avoidance tests such as acqusition trial and retention test in DHA group were significantly higher (42% and 15%, respectively) than those in control group. Acetylcholinesterase (AChE) activity was significantly increased about 15% by long-term administration of DHA-rich fish oil compared with control group. Neurotransmitters such as dopamine (DA) and serotonin (5-HT) in DHA group were significantly about 15% lower than those in control group, whereas their metabolites such as homovanillic acid (HVA) and 5-hydroxyindole acetic acid (5-HIAA) in DHA group were significantly higher (30% and 20%, respectively) than those in control group. Therefore, the ratios of HVA/DA and 5-HIAA/5-HT as a ratio of metabolites on neurotransmitter were significantly higher (50% and 40%, respectively) than those in control group. These results suggest that long-term administration of DHA-fish oil may be activated retention test, acetylcholinesterase activity, and ratios of metabolite on neurotransmitter, resulting in greater prevention of learning and memory impairments such as Alzheimer-type dementia.
