Expression of Alzheimer¡¯s Amyloid Precursor Protein (¥âAPP) in the Brain of Senescence Accelerated Mouse (SAM)

Patients with Alzheimer¡¯s disease (AD) show the loss of memory and the cognitive deficits of which molecular mechanisms were exactly not known. We examined age-related changes in the expression levels of ¥â-amyloid precursor protein (¥âAPP) in the brain of senescence accelerated mouse (SAM)-P/1, which shows the senile amyloidosis and the impairment of hearing, and SAM-, P/10, which shows age-dependent brain atrophy and impairment in learning and memory, using Northern blot hybridization, reverse transcription coupled polymerase chain reaction (RT-PCR) and western blot. ¥âAPP695 mRNA is only expressed in the brains of SAM. The changes of the amount of APP mRNA in the presencephalon and the mesencephalon are observed significantly with advancing age using Northern blotting. However the expressions of ¥âAPP were changed with aging in only hippocampus of SAM-P/10 in the level of mRNA and protein. Moreover, ¥âAPP695 was reached a peak level at 12 months of age in the procencephalon and mesencephalon of SAM-P/1. ¥âAPP mRNA and protein were found to be reached a peak level at 8 months of age in the hippocampus of SAM-P/10. These results suggest that the ¥âAPP overexpression in the brain may contribute to the aging of SAM-P/1 and the changes of ¥âAPP in the hippocampus might be related to the characteristic memory loss in SAM-P/10.
