Opuntia Ficus-Indica Ethanol Extract Ameliorates Streptozotocin-Induced Hyperglycemia in Rats

Abstract
Opuntia ficus-indica ethanol extract(OFEE) has been known to possess several therapeutic properties, such as an anti-inflammatory and hypoglycemic actions in Oriental medicine. However, little is known about its exact pharmacological mechanism on the regulation of blood glucose in normal and diabetic state. To evaluate the effect of OFEE on the regulation of blood glucose, we made streptozotocin(STZ)- induced diabetic rat model, and examined whether OFEE treatment can exhibit the hypoglycemic effect in these animals. STZ(50 mg/kg, i.v.)-treated rats showed the hyperglycemia (blood glucose changed from 150 mg/dl to over 400 mg/dl) within 3 days after its injection. The OFEE administration significantly (p<0.05) reduced the blood glucose level from 400 mg/dl to 300 mg/dl at 4 hours, but it did not affect at level in normoglycemic rats. On the other hand, a remarkable reduction of pancreatic beta cells and insulin(+) cells was exhibited in STZ-induced hyperglycemic rats by immunohistochemical analyses. We also examined whether OFEE has an alleviating effect on the damage of pancreatic beta cells or insulin(+) cells induced by STZ, and failed to observe a protective effect of OFEE on the islet cell and its insulin(+) cells of pancreas. Our results suggest a possibilty that the hypoglycemic action of OFEE is due to other mechanism, than the protection of the insulin(+) beta cells in STZ-induced diabetic rats.
