Mitochondrial Dysfunction and Oxidative Stress in Cytotoxicity Induced by Amyloid Peptide

Abstract

|
Recent studies suggest that oxygen free radicals are involved in the cytotoxic effects of amyloid-¥â-peptide (A¥â). In this study, we investigated the induction of oxidative stress and mitochondrial dysfunction in the process of cell death induced by A¥â peptides. A¥â treatment to the C6 cell increased levels of DCF-fluorescence, suggesting an increased intracellular concentration of hydrogen peroxide and furthermore an oxidative stress in cells. Activity of antioxidant enzyme catalase and superoxide dismuatase showed no significant difference between control and A¥â-treated C6 cell. The activities of mitochondrial enzymes, succinate dehydrogenase(SDH) and cytochrome C oxidase(COX) decreased by the treatment of A¥â to the C6 cells. Pretreatment of antioxidant eliminate the reductive effect of A¥â on SDH and COX activity. In the parallel experiment the A¥â-treatment also induced LDH-release, which indicates cytotoxicity. However antioxidant pretreatment did not block the reduction of LDH-release in A¥â-exposed C6 cell. liken together, our results suggest that the oxidative stress associated with mitochondrial dysfunction induced by A¥â.
