Effect of Iron Accumulation on Mitochondrial Permeability Transition and Oxidation of Rat Heart Mitochondria

|
Oxidation level and mitochondrial permeability transition(MPT) of mitochondria were investigated in iron-injected rat heart. There were increase in lipid peroxidation(MDA), free radical(DCFDA), and protein oxidation(carbonyl content) by 227%, 82%, and 18%, respectively in iron-injected mitochondria. Thiol group was decreased by 24% in iron-overloaded mitochondria. Iron and calcium contents in mitochondria were also increased by 35% and 112%, respectively in iron-injected rats. This result indicates that iron accumulation in mitochondria may be responsible for the increased oxidation and calcium content. In Iron-injected rat, MPT was reduced. However, ferrous citrate directly added to heart mitochondria raised the MPT. This opposite effect of iron suggests the MPT dependence on iron location, which may be related with reversible oxidation of membrane components.
