Abstract
Carlic has been used as food and as medicine in many parts of tile world for centuries. It has been well known to have antibacterial activity, lipid-lowering efffcts, anti-platelet-aggregation. drug metabolizing properties, anticarrinogenic artions, and other medical effects. In this study, aqueous extract of garlic and one if its major component allyl suIfide were used to examine their cytotoxic effect on human cancer cell lines of carious origins together with two normal cells. The types ofcancercell lines used in this study are SK-Hep1, Hep3B, HeLa, SDK, G-292, KHOS/NP and MG-63. Two normal cells are human gingival tissue cells mostly consisted of fibroblasts and immortalized human liver cells whose characteristics are in between normal and cancer cells. The effect of adriamycin, known as one of most potent anticancer drog widely used was compared with that of garlic. THe [3-(4,5-dimethyl thiazol-2yl), 2-5-diphenyl-tetrazo-lion bromide] assay was employed to check the cytotoxicity.
The results obtained were as follows:
1. Aqveous extract of garlic showed a considerable cytotoxic effect on immortalized liver cells, while it didn¢¥t inhibit growth of the normal gingival tissue cells.
2. The cytotoxic effect of aqueous extract of garlic on cancer tell lines was markedly different depending on The origins of cancer, among which, SK-Hep1 was most sensitively responded.
3. Though allyl sulfide, a major component of garlic brought more inhibition to SK-Hep1 than aqueous extract of garlic, it also inhibited normal cell growth.
4. When adriamycin was treated on cancer cell lines with the concentrations of 9-46 uM, 30-85% inhibition of cell growth was observed. This cytotoxic effect of adriamycin was more potenciated by combined use of aqueous extract of garlic in all cancer cell lines used.
