Effect or Morphine and Gabapentin on the Mechanical Allodynia in the Spinal Cord Hemisected Rats

|
Spinal cord injury patients may suffer from chronic central pain, which is difficult to treat with conventional analgesics. However, in animal studies, it has been reported that morphine reduced the signs of spinal cord injury-induced pain. Probably, this disparity between the two results seems to be due to the difference in a point of time of treatment. To test this hypothesis, we performed to compare the efficacy of morphine and gabapentin between early phase and late phase following spinal cord injury. Spinal cord injury was made by hemisection of the spinal cord at the level of T13 in male Sprague-Dawley (150-200 g) rats under enflurane anesthesia. Mechanical allodynia was assessed by the response frequency of hindpaw withdrawal to von Frey hair (2.0 g) stimulation applied to the plantar surface. In order to compare the effect of morphine and gabapentin on mechanical allodynia between early phase (3 weeks after injury; 3 W group) and late phase (16 weeks after injury; 16 W group) following hemisection, morphine (2 mg/kg), and gabapentin (30 mg/kg) were injected intraperitorneally and behavioral test were conducted in each group. Antiallodynic effect of morphine was much more pronounced in the 3W group than in the 16 W groups. However, the relieving effect of gabapentin was not significantly different between the 3 W and the 16 W group. These results suggest that the efficacy of morphine on the mechanical allodynia is alleviated according to the lapse of time following spinal cord hemisection, but the efficacy of gabapentin does not change.
