ALTERATION OF COLONIC MOTILITY IN TYPE 2 DIABETES MELLITUS MODEL IN RAT

Abstract

|
It is reported that approximately 45% of patients with diabetes mellitus have gastrointestinal complications such as diarrhea and constipation. However, the cause of disordered motility is unclear. The present study investigates alterations in colonic motility that may be responsible for these symptoms in vitro and in vivo using an established animal model of type 2 diabetes mellitus. A type 2 diabetes mellitus model in adulthood was produced by intraperitoneal administration of streptozotocin to neonatal rats. Rats were sacrificed after 8 weeks and distal colonic muscle strips were prepared. Responses of these strips to neurotransmitter agents and electrical field stimulation (EFS) were recorded in vitro. The contractile responses to high K+ and carbachol or to EFS was not different. Yet EFS-induced contraction was significantly decreased under atropine (1¥ìM), whereas it was significantly increased under N -nitro-L-arginine methylester (L-NAME; 100¥ìM) in diabetic colon compared to normal colon. in vivo study, bethanechol (BCh) and L-NAME were administered for measurement number of fecal output. Bethanechol (1.5 mg/kg) significantly increased fecal output in diabetes rats compared to control rats. L-NAME (10 mg/kg) significantly decreased bethanechol-induced fecal output in diabetes rats, but slightly reduced in normal rats. These results demonstrate that there is a marked alteration in motility in the colon of diabetic rats. The change in motility is due to both an increased cholinergic and nitrergic responses.
