Propofol Potentiate the Neuromuscular Blocking Effect of Mivacurium in the Rat Hemidiaphragm

Abstract
INTRODUCTION: It has been reported that propofol of high concentration potentiates the neuromu-scular blocking effect of vecuronium in the rat hemidiaphragm1). We have evaluated the inter-action between mivacurium and propofol in the rat isolated phrenic nerve-hemidiaphragm.

METHOD: Phrenic nerve was stimulated in a train-of-four fasion. Dose-response curves for propofol alone(n=12, P1 group), mivacurium alone(n=12, M1 group), mivacurium in the presence of maximum propofol concentration showed no effect on twitch tension(n=12, M12 group) and propofol in the presence of mivacurium ED50(n=12, P2 group) were obtained by increasing the concentration of mivacurium 0.5 mulol liter 1 or propofol 100 mulol liter 1 in the rat hemidiaphragm. Comparison of the ED50 value for M1 group were performed using analysis of variance and Student¢¥s t test.

RESULT: Propofol concentration of 100 mulol liter 1 was the maximum concentration that showed no effect on twitch tension in P1 group. Propofol inhibited twitch tension in a concentration dependent manner. The mivacu-rium concentration required to inhibit T1 by 50%(ED50) was 2.633+/-0.11 mulol liter-1 in M1 group and 2.16+/-0.12 mulol liter 1 in M2 group. The mivacurium concentration required to inhibit T4 by 50%(ED50) was 2.03+/-0.09 mulol liter 1 in M1 group (Fig. 1.). In each case the reduction in ED50 was stastistically significant(P < 0.05), Propofol concentration of 100 mulol liter 1 showed no twitch inhibition in P1 group but significant twitch inhibition in P2 group.

CONCLUSION: Propofol potentiate neuromuscular blocking effect of mivacurium in rat hemidiaphragm.
