A Study of Biological Mechanism of Antipsychotics-Induced Weight loss

Abstract
Topiramate is a new anticonvulsant, which is being investigated as a mood stabilizer, with weight loss as a frequent side effect. The mechanism of this is presently unknown, but most patients express this as a desirable side effect. In the present study, the effects of topiramate on expression of leptin and ob-Rb mRNA in 3T3-LI cell line was studied. For that purpose, the method of RT-PCR was applied. In 3T3-LI cells treated with topiramate, the level of leptin and ob-Rb mRNA were slightly increased as compared as control cells. These results suggest that mechanism of weight loss of topiramate is through induction of leptin and ob-Rb activity. Also, this study would provide supporting data on the possibility of the involvement of leptin and its receptor in the mechanism of topiramate-induced weight loss.
