The Effects of UCP Polymorphisms on Obesity Phenotypes and Weight Loss among Korean Overweight Women

Abstract
Objectives :This study was conducted to investigate the effetcs of UCP(uncoupling protein) 1-4 polymorphisms on obesity and weight loss. For this, we analyzed association between polymorphisms of UCP 1-4 and phenotypes of obesity(BMI, percent-body-fat, and abdominal fat area) and changed percentage of phenotypes(BMI and percent-body-fat) after treatment.

Methods :A total of 425 Korean women were recruited from obesity clinic of Kirin Oriental Medical Hospital(Seoul, Korea). All subjects were BMI over 25. Among total subjects, 207 females treated with VLCD(very low calorie diet) and oriental medical therapy for a month were analyzed that UCP phenotype influenced on change of obesity-phenotypes. For genotyping of SNPs, genomic DNA from each subject was extracted from whole blood and genotyped by TaqMan Method. Haplotypes and their frequencies were inferred using algorithm, HapAnalyzer Program. BMI, percent-body-fat, and total-subcutaneous-visceral abdominal fat area were measured. Associations among those values, polymorphisms, and haplotypes of UCPs were analyzed by age-adjusted general linear model.

Results and Conclusions:In this study, GG type of UCP-1 -1766A>G, AG, GG type of UCP-2 -866A>G, CC, CT type of UCP-2 +4787C>T, CG, GG type of UCP-3 +2564C>G, AG, GG type of UCP-3 +3106A>G, TC, CC type of UCP-3 +4589T>C, and -/ht, ht/ht type of UCP2-3 ht1 were associated with obesity and played a role as a resistance gene of weight loss.
