^(3)H-imipramine Receptor Assay in Rat Brain Tissues

Abstract

|
In synaptic transmission the catecholamine and it¡¯s ©øH-imipramine bindings stimulate the adenylate cyclase activity which reflect the change of biological response. In rat medicated with the agonist to activate metabolites, the change of catecholamines and it¡¯s ©øH-imipramine binding sites in the nerve system were as follows. l. In rat brain tissues, norepinephrine level was 370¡¾10.8 ng/g, epinephrine was 1107.2 ng/g¡¾7.2 ng/g, dopamine was 810¡¾0.26 ng/g, ©øH-imipramine receptor was 23.7+3.2 fmol/rm of protein, affinity(Kd) was 3.8¡¾0.261 nmol. 2. In administrated with imipramine to affect as agonist to activate adrenergic receptor in the rats and the depression patients, the plasma concentrations of circulating catecholamine were increased. Such results shows that the low plasma levels of the norepinephrine could be increased in the disease process of depression patients medicated with imipramine as agonist to activate metabolites, and so imipramine medication could be increased the serum levels of norepinephrine and it¡¯s 3H-imipramine receptor in the rat brain tissues.
