Effect of Lipids Containing Conjugated Linoleic Acids on Proliferation and Differentiation of Preadipocytic Cells

Conjugated linoleic acid (CLA) have been recently reported to have an anti-obesity effect in animals and humans. The objectives of this study were to investigate the anti-obesity effect of lipids containing CLA in 3T3-L1 preadipocytes. Cell viability was determined using WST-8 analysis and cell differentiation was determined by glycerol-3-phophate dehydrogenase (GPDH) activity. Lipid accumulation in differentiating 3T3-L1 cells was measured by Oil red O staining There were six experimental groups including vehicle control (DMSO), diglyceride (DG), CLA, triglyceride (TG)-CLA, DG+CLA20, and DG+CLA60. The mean viability of 3T3-L1 cells by treatments of test solutions at the concentrations of 10 to 1000 §¶/ml was reduced in a dose-dependent manner. The test solutions showed a similar pattern in reduction of cell viability. Treatments of the test solutions at the concentrations of 500 ¥ìg/ml for 48 h in 3T3-L1 cells inhibited the differentiation of as measured by GPDH activity after incubation in adequate differentiation media for 8 days. In addition, lipid accumulation was also reduced as incubated for 8 days in differentiation medium after treatments of test solutions for 48 h in 3T3-L1 cells. Among them DG+CLA60 was the most effective in the inhibition of differentiation and lipid accumulation. These results suggest that the compounds including CLA as fatty acids may have respectable anti-obesity effects, regardless ofdiglyceride and/or triglyceride or CLA alone and that they may give a benefit for controlling overweight in humans.
