Optimization of Enzyme Membrane for a Needle - type L - Lactate Biosensor

Abstract

An enzyme membrane compatible with a needle-type compound H©üO©üelectrode, composed of platinum working and Ag/ AgCl reference electrodes, was prepared and the corresponding L-lactate biosensor using the membrane was evaluated for electrochemical properties. The biosensor having the enzyme-containing layer only showed a sigmoidally increasing current response against substrate addition and an inhibition current owing to L-ascorbic acid oxidation. Potential dependence of biosensor response was also evident from cyclovoltammetric analysis. In the biosensor having the enzyme-containing layer only, the time required to reach steady-state current response was 1.8¡3.0 min. L-Ascorbic acid interference was conspicuously removed (over 91¡98%) by inner layer coatings with Nafion, cellulose acetate and ethyl cellulose. By outer layer coatings with ethyl cellulose and cellulose acetate, the dynamic range of the biosensor increased by more than ten-fold, compared to that of the biosensor without the outer layer.
