Liquefaction Methods for Production of Sulfated Mucopolysaccharides from Ciona intestinalis

Abstract
The effect of liquefaction methods on the content of sulfated mucopolysaccharides (MPS) in Ciona intestinalis was studied. The extracts by pressure cooker at 121¡É and 1.5 §¸/§² had the highest content of sulfated MPS (8.6-10.3%), followed by those treated with boiling water (4.3-5.9%). Acid/alkali and enzyme treatments produced lower contents of sulfated MPS than extracts by the pressure cooker. Treatments of Alcalase and Neutrase gave higher contents of sulfated MPS (5.4% and 3.7%, respectively) than other enzyme treatments. Compared with all liquefaction methods, pressure cooking for 1 hr was effective for the preparation of sulfated MPS products, and its recovery of sulfated MPS was approximately 50%.
