The Effects of Anthocyanins Isolated from Vitis Coignetiae Pulliat on NF-?B in Human Cancer Cells

Abstract
NF-?B activation pathway is involved in the cancer cell proliferation, invasion, angiogenesis, metastasis, and drug resistance. The previous studies suggested that anthocyanins from the Fruit of Vitis Coignetiae Pulliat (Meoru in Korea, AIMs) might suppressed NF-?B activation in cancer cells. Here, we investigated the effects of AIMs on NF-?B activity in human cancer cell lines (HT-29, HeLa, and Hep3B). We found that AIMs inhibited NF-?B activity of all the three cancer cell lines. This finding was confirmed by Western blot and luciferase activity assays in HT-29 cells. The AIMs inhibited the invasion of HT-29 cells and also inhibited the expression of MMP-2 and MMP-9 which are key molecules in cancer cell invasion. We also confirmed this finding in HeLa cells. Taken together, this study indicates that AIMs have inhibitory effects on NF-?B in human cancer cell lines HT-29, HeLa, and Hep3B. This suppression of NF-?B activity led to inhibition of cancer cell invasion through suppression of MMP-2 and MMP-9 expression.
