Antioxidant Effect of Spermine Is Related to Inhibition of Matrix Metalloproteinase in SW480 Cells

Polyamines such as putrescine, spermidine and spermine are low molecular weight cations present in mammalian cells. The effect of spermine was investigated on antioxidant and matrix metalloproteinase-9 (MMP-9) in human colon cancer cells, SW480 cells. Thus, first of all, the direct scavenging effect of spermine on DPPH radical, superoxide anion, hydroxyl radical, hydrogen peroxide, reducing power and protective effect of DNA oxidation related to oxidative stress were evaluated in cell free system. In addition, its inhibitory effects on the activity and expression of MMP-2 and -9 related to metastasis were determined using gelatin zymography. In this study, it was observed that spermine exhibited higher inhibitory effects on hydrogen peroxide and hydroxyl radical than those of reactive oxygen species, and blocked DNA oxidation induced by hydroxyl radical. Moreover, it specifically inhibited both activity and expression of MMP-9 in SW480 cells. These results suggest that inhibitory effect of oxidative stress in the presence of spermine could inhibit the expression of MMP-9 in SW480 cells.
